| 目的研究二烯丙基二硫（DADS）是否通过下调组织蛋白酶D（Cathepsin D）的表达抑制人胃癌MGC803细胞侵袭转移。方法RT-PCR和Western blot检测DADS及干扰Cath-D对人胃癌MGC803细胞Cath-D mRNA与蛋白表达变化；细胞划痕实验和Transwell实验分别检测DADS及干扰Cath-D对人胃癌MGC803细胞侵袭转移能力的变化。结果DADS作用人胃癌MGC803细胞24 h后，Cath-D mRNA与蛋白表达水平明显下降(P＜0.05)，划痕距离明显增宽(P＜0.05)，穿膜细胞数明显减少(P＜0.05)；siRNA干扰Cath-D后，Cath-D mRNA与蛋白的表达水平明显下降(P＜0.05)，划痕距离明显增宽(P＜0.05)，穿膜细胞数明显减少(P＜0.05)。结论DADS能抑制人胃癌MGC803细胞的侵袭转移能力，其机制可能与下调Cath-D 的表达有关。 |

| ObjectiveTo investigate whether diallyl disulfide (DADS) inhibits human gastric carcinoma MGC803 cells invasion and metastasis by down-regulating the expression of Cathepsin D.MethodsThe mRNA and protein expression level of Cath-D were detected by RT-PCR and Western blot respectively;Migration and invasion potentials were examined by Wound Healing assay and transwell assay.ResultsHuman gastric carcinoma MGC803 cells exposed to DADS30 mg/L for 24h,the mRNA and protein expression level of Cath-D were significantly decreased (P<0.05),scar distance was widened (P<0.05),the number of cancer cells which passed through the Matrigel coated membrane was reduced (P<0.05);after Cath-D was interfered by siRNA,the mRNA and protein expression level of Cath-D were significantly decreased (P<0.05),scar distance was widened (P<0.05),the number of cancer cells which passed through the Matrigel coated membrane was reduced (P<0.05).ConclusionsDADS can significantly inhibit the ability of migration and invasion ability of human gastric carcinoma MGC803 cells and its mechanism may be related to the expression of down-regulating Cath-D. |
