目的研究二烯丙基二硫(DADS)对人髓系白血病细胞株HL-60细胞增殖及对c-myc基因表达的影响。方法采用MTT和细胞记数法分析DADS对细胞生长的影响、蛋白印迹法检测细胞内c-myc蛋白表达水平。结果 DADS能抑制HL-60细胞增殖,并呈剂量和时间依赖性(P<0.05),DADS(20 μmol/L)作用12 h后能下调c-myc蛋白的表达水平。结论DADS抑制HL-60细胞增殖,其机制可能与下调c-myc表达水平密切相关。 |

ObjectiveTo investigate the effects of DADS on proliferation inhibition and on c-myc protein expression in human myeloid leukemia cell line HL-60 cells.MethodsHL-60 cells were exposed to DADS at different dosages.Growth inhibition was detected by MTT assay and cell growth.The expression of c-myc was measured by Western blotting.ResultsAfter treatment with DADS, the growth of HL-60 cells were suppressed in a concentration-dependent and time-dependent manner(P<0.05).Incubation of HL-60 cells with DADS(20 μmol/L)for 12 h could decrease the expression of c-myc.ConclusionDADS could inhibit HL-60 cells proliferation,its mechanism may be closely related with adjusting expression level of the c-myc. |
