      目的探讨辛二酰苯胺异羟肟酸(SAHA)对转化生长因子-β1(TGF-β1)所诱导的人胚肺成纤维细胞(HELF)胶原表达的影响。方法体外培养人胚肺成纤维细胞系HELF,并分为空白对照组、SAHA组、TGF-β组以及SAHA+TGF-β组。其中空白对照组加入等体积PBS;SAHA组加入5 μmol/L SAHA;TGF-β组加入终浓度为5 ng/mL TGF-β1孵育24 h;SAHA+TGF-β组在TGF-β组基础上分别加入0.5、2.0、和5.0 mol/L SAHA,共同孵育24 h。处理结束后,获取培养上清,比色法测量羟脯氨酸(HYP)的含量,并用逆转录-聚合酶链反应(RT-PCR)检测HELF细胞中Ⅰ、Ⅲ型前胶原的mRNA表达水平。结果对照组脯氨酸表达极低,TGF-β1处理后,羟脯氨酸含量达12.684±1.485 μg/mL。当分别用0.5、2.0、和5.0 mol/L SAHA处理后,HYP含量随着SAHA浓度的增加而减少(P<0.05);SAHA孵育24 h后,能够明显抑制TGF-β1刺激后的细胞表达Ⅰ型和Ⅲ型前胶原蛋白mRNA,并且具有明显的剂量依赖性(P<0.05)。结论SAHA能减少TGF-β1诱导的HELF羟脯氨酸含量及Ⅰ、Ⅲ型前胶原蛋白表达,这可能是其抗纤维化的重要机制。

      ObjectiveTo investigate the effect of suberoylanilide hydroxamic acid (SAHA) on transforming growth factor-β1 (TGF-β1)-induced collagen protein expression in human embryonic lung fibroblast cells (HELF).MethodsHELF cells were cultured in vitro and divided into Control group (group A),SAHA group(group B),TGF-β group (group C) and SAHA groups (group D) according to the different purpose.PBS was used as negative control,and the cells were stimulated with 5 ng/mL of TGF-β for 24h.For group B,5mol/L SAHA was added.In group C,5 ng/mL of TGF-β was used.HELF cells in group D were treated with 5 ng/mL of TGF-β followed by SAHA at 0.5mol/L ,2mol/L or 5mol/L for an additional 24h.Concentration of Hyfroxyprotein was determined by colorimetry,and the mRNA levels of I,Ⅲ procollagen in HELF cells were detected by RT-PCR.ResultsHydroxyproline expression was very low in the control group,while the hydroxyproline content reached to (12.684±1.485) μg/mL after TGF-β1 treatment.When HELF cells were treated with 0.5 μM,2 μM or 5 μM SAHA,hydroxyproline content decreased with the increase of concentration of SAHA (P<0.05).After 24h incubation of SAHA,mRNA expression of type I and type Ⅲ procollagen can be inhibited by stimulation of TGF-β1 in dose-dependent manner.ConclusionsSAHA can inhibit TGF-β1-induced Hyfroxyprotein production,I and type Ⅲ procollagen expression,and this may be one of the anti-fibrosis mechanisms.
