| 目的探讨槟榔碱对3T3-L1前脂肪细胞增殖、 分化的影响及其机制。方法采用四甲基偶氮唑盐（MTT）法和油红 O染色检测不同浓度（10、30、50、100 μmol /L）的槟榔碱对3T3-L1前脂肪细胞增殖和分化的影响，荧光定量PCR检测过氧化物酶体增殖物激活受体γ（PPAR γ）和CAA T/增强子结合蛋白α(C/ EBP α)的表达。结果10～100 μmol /L 槟榔碱作用 3T3-L1前脂肪细胞12 h和24 h能促进3T3-L1前脂肪细胞增殖，且呈一定的量效关系。到分化的第9天，与对照组相比，50 μmol /L 槟榔碱组胞浆中的脂滴明显减少，同时PPAR γ和C/ EBP α mRNA表达水平显著降低。结论槟榔碱能促进3T3-L1前脂肪细胞的增殖、抑制其分化，其机制可能与PPARγ和C/ EBPα的表达降低有关。 |

| objectivesTo investigate the effects of arecoline on the proliferation and differentiation of 3T3-L1 preadipocytes and to elucidate its possible mechanism.Methods3T3-L1 preadipocytes were cultured and treated with different concentrations of arecoline （10、30、50、100 μmol /L），the proliferation and differentiation of 3T3-L1 preadipocytes were evaluated by MTT methods and oil red O staining，respectively.The expressions of peroxisome proliferation activated receptor γ (PPAR γ) and CAAT/ enhancer binding protein (C/ EBP α) mRNAs were detected by quantitative real-time RT-PCR (qRT-PCR).ResultsTreatment of arecoline at different concentrations (10～100 μmol /L) for 12 and 24 h significantly promoted 3T3-L1 preadipocyte proliferation in a dose-depend manner.On the day 9 of differentiation，compared with the control group，lipid droplets in the cytoplasma and the PPARγ and C/EBP α mRNA expression were significantly decreased in the 50 μmol /L arecoline group.ConclusionArecoline promotes the proliferation and inhibits the differentiation of 3T3-L1 preadipocytes，which may be associated with decreased expressions of PPAR γ and C/ EBP α mRNA. |
