目的对非小细胞肺癌(NSCLC)组织中血管内皮细胞生长因子C(VEGF-C)、VEGF-D及基质金属蛋白酶-12(MMP-12)的表达进行检测,探讨这3个因子与NSCLC纵隔淋巴结转移的相关性。方法采集60例NSCLC组织和癌旁组织,应用免疫组织化学方法检测其VEGF-C,VEGF-D及MMP-12的表达,并分析其与临床病理因素之间的相关性。结果VEGF-C,VEGF-D及MMP-12蛋白在NSCLC组织中呈高表达(96.7%、93.3%及91.7%),显著高于癌旁组织(P<0.01)。 VEGF-C,VEGF-D及MMP-12蛋白的表达在有淋巴结转移的病例显著高于没有转移的病例(χ2=13.237,χ2=6.960,χ2=9.870;P=0.000,P=0.008,P=0.002)。VEGF-C,VEGF-D及MMP-12蛋白表达与性别、年龄、分化程度以及组织学类型无关(χ2=0.472;P=0.244),但与病理TNM分期(PTNM)有关(P=0.01)。在有纵隔淋巴结转移的组织中,VEGF-C,VEGF-D与MMP-12蛋白表达均呈正相关(r=0.850,P=0.000;r=0.787,P=0.000;r=0.914,P=0.000)。结论VEGF-C,VEGF-D及MMP-12蛋白在有纵隔淋巴结转移NSCLC的高表达与其淋巴结转移密切相关,VEGF-C,VEGF-D及MMP-12蛋白有成为NSCLC治疗中新靶位点的潜在可能。 |

ObjectiveThrough detecting the expressions of VEGF-C, VEGF-D and MMP-12 in the tissues of Non-small cell lung cancer and peritumoral, to investigate their correlations with lymph node metastasis of NSCLC.Methods Expressions of VEGF-C, VEGF-D and MMP-12 in 60 cases of tissues of Non-small cell lung cancer and peritumoral were detected by immunohistochemistry, and their correlations with clinical-pathological findings were analysed in patients.ResultsThe positive rates of VEGF-C,VEGF-D andMMP-12 were 96.7%%,93.3%,91.7% respectively,which were significantly higher than those of the tissues of peritumoral(P<0.01).The expression of VEGF-C,VEGF-D and MMP-12 protein in lymph node metastasis was significantly higher than the cases without lymph node metastasis(χ2=13.237,χ2=6.960,χ2=9.870;P=0.000, P=0.000, P=0.000).The expressions of VEGF-C,VEGF-D and mmp-12 protein in NSCLC were not correlated with gender,age,degree of differentiation and histological types(χ2=0.472;P=0.244),but correlated significantly with PTNM stage(P=0.01).Significant correlations between VEGF-C and VEGF-D(r=0.929,P=0.000),VEGF-C and MMP-12(r=0.787,P=0.000),VEGF-D and MMP-12(r=0.914,P=0.000) expression were observed in LNM.ConclusionsThere was significant correlation between the high expression of VEGF-C,VEGF-D and MMP-12 and lymph node metastasis in tissues of NSCLC. |
