      目的探讨金樱子对链脲佐菌素(STZ)诱导的糖尿病大鼠肾脏氧化应激反应的影响,并探讨其作用机理。方法STZ腹腔注射诱导建立SD(Sprague-Dawley)大鼠糖尿病模型后,随机分为糖尿病模型组和金樱子干预组,同时另设正常对照组和金樱子对照组。测定大鼠肾组织总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。免疫组织化学、Western blot,Real-time PCR检测各组肾组织核转录因子κB(NF-κB)表达情况。结果与正常对照组相比,糖尿病模型组大鼠肾组织T-AOC,SOD活性明显减弱,MDA含量明显增强(P<0.01),而糖尿病大鼠经金樱子处理后,肾组织T-AOC,SOD的活性显著增强,MDA含量显著降低(P<0.05),且能在mRNA和蛋白水平显著下调肾组织中NF-κB的表达。结论金樱子可通过降低糖尿病大鼠肾脏中NF-κB表达,抑制氧化应激,增强抗氧化酶活性起到保护肾脏的作用。

      ObjectiveTo investigate the effect of Rosa Laevigata Mickx(RLM) on oxidative stress in diabetic nephropathy rats,and explore the mechanism of action.MethodsThe Sprague-Dawley (SD) rats induced by STZ were divided into the diabetic model group and the RLM intervention group,and the normal control group and the RLM control group were set up. The ability of anti-oxidant capacity (T-AOC),the activity of superoxide dismutase (SOD),the amount of malondialdehyde (MDA) in renal tissues were determined. The expression of nuclear factor-kappa B (NF-κB) in the four groups were detected by immunohistochemistrcey,Western blot and real-time PCR. ResultsCompared to the normal group,the activity of SOD and T-AOC were significantly decreased,and the level of MDA were significantly increased in the diabetic rats (P<0.01). However,RLM significantly increased the activity of SOD and T-AOC,decreased the level of MDA (P<0.05),and inhibited the expression of nuclear factor-kappa B (NF-κB) at both the protein and mRNA levels.ConclusionsRosa Laevigata Mickx. can inhibit the oxidative stress,increase the activity of antioxidase,decrease the oxidative damage of diabetic nephropathy,thereby protect the kidney in diabetic,which may be related to decreasing the expression of NF-κB.
