目的观察磷脂酰肌醇-3-激酶的特异性抑制剂(LY294002)对胃癌MGC803细胞生长的影响。方法选用LY294002处理胃癌MGC803细胞后,利用细胞生长曲线、平皿克隆与流式细胞术观察细胞的生长情况,采用免疫细胞化学染色和Western blot检测AKT蛋白与p27Kip1蛋白磷酸化及Cyclin E蛋白的表达。结果LY294002处理后,MGC803细胞生长速度明显减慢,细胞克隆体积变小,数量减少;G1期细胞比例明显增加,S期细胞比例降低。LY294002处理后,MGC803细胞中磷酸化的AKT蛋白与Cyclin E蛋白表达降低,而磷酸化的p27Kip1蛋白表达升高。 结论LY294002抑制胃癌MGC803细胞生长速度,呈现出G1/S期细胞周期阻滞。 |

ObjectivesTo observe the growth of MGC803 cells affected by LY294002 (Specific inhibitor of phosphatidylinositol -3-kinase).MethodsThe growth and cell cycle of MGC803 cells, treated by LY294002, were observed by draw cellular growth curve and plate clone formation and flow cytometry. The expressions of phosphorylation of AKT protein and p27Kip1 protein and of CyclinE protein were detected by immuocytochemistry and Western blot in MGC803 cells.ResultsAfter LY294002 treated MGC803 cells, the cellular growth velocity obviously depressed and the volume of cell clone shrinked and its quantity decreased; The G1 period rate of MGC803 cells gradually raised and S period obviously decreased. The expression of phosphorylation AKT protein and Cyclin E protein down-regulated, but the phosphorylation p27Kip1 protein decreased, after MGC803 cells treated by LY294002. ConclusionThe cellular growth velocity of gastric carcinoma MGC803 cells treated by LY294002 was degraded, and MGC803 cells were held-up in cell cycle of G1/S period. |
