      目的研究油酸(OA)对小鼠c2c12细胞中过氧化物酶体增殖物激活受体γ辅激活因子1α(PGC-1α)表达的影响,探讨p38丝裂原活化蛋白激酶(MAPK)信号通路在其中的作用。方法以不同浓度油酸及p38抑制剂SB203580(SB)分别处理c2c12细胞, 逆转录-聚合酶链反应(RT-PCR)检测PGC-1α mRNA表达, Western Blot检测PGC-1α蛋白表达。结果油酸处理组PGC-1α mRNA及蛋白表达明显高于对照组(P<0.05); OA+SB组PGC-1α表达减低(P<0.05)。结论油酸能促进c2c12细胞PGC-1α mRNA转录和蛋白的表达,该作用可能与p38 MAPK信号通路有关。

      ObjectiveTo investigate the effect of oleic acid(OA) on the expression of PGC-1α mRNA and protein in mouse c2c12 cells and to unravel the mechanism of its transcriptional regulation. MethodsCultured c2c12 cells were treated by oleic acid with or without SB203580(SB) respectively; PGC-1α mRNA was detected by RT-PCR; PGC-1α protein were detected by Western Blot. ResultsCompared with control group, the expression of PGC-1α mRNA and protein increased significantly in OA groups (P<0.05); compared with the OA group, the expression of PGC-1α mRNA and protein decreased significantly in OA+SB group (P<0.05). ConclusionOleic acid can stimulate the expression of PGC-1α mRNA and protein in c2c12 cells; oleic acid might regulate the PGC-1α gene expression through p38 MAPK pathway.
