      目的探讨过表达CAMP反应元件结合蛋白(CREB)在内皮细胞氧化损伤中的作用。方法建立H2O2诱导的血管内皮细胞损伤模型,将内皮细胞ECV304分为对照组、H2O2组、转染PCI空载体组、转染PCI空载体+H2O2组、转染PCI-CRESAP组和转染PCI-CREB+H2O2组,采用MTT法检测细胞存活率,硫代巴比妥酸检测丙二醛(MDA)含量,黄嘌呤氧化酶法检测细胞超氧化物歧化酶(SOD)活性。结果转染CREB+H2O2组的细胞存活率和SOD活性明显高于H2O2组(P<0.05),而转染CREB+H2O2组的MDA含量明显低于H2O2组(P<0.05)。结论过表达CREB对H2O2诱导的内皮细胞氧化损伤具有保护作用。

      ObjectiveTo investigate the effect of overexpression of CREB on oxidative injury in endothelial cells.MethodsThe endothelial cell oxidative injury model was induced by H2O2.ECV304 cells were divided into 6 groups:control group,H2O2 group,PCI transfection group,PCI transfection+H2O2 group,PCI-CREB transfection group and PCI-CREB transfection+H2O2 group.The viablity of ECV304 cells was detected by MTT.The MDA concentration was observed by TBA method.Levels of SOD were tested by xanthine oxidase.ResultsCompared with H2O2 group,both cell viablity and SOD activity in PCI-CREB transfection+H2O2 group showed a significant increase,and MDA concentration decreased (P<0.05).ConclusionOverexpression of CREB has protection role during oxidative injury induced by H2O2.
