目的探讨体外培养条件下高糖状态对人脐静脉内皮细胞假性血友病因子(von Willebrand factor,vWF)、内皮素-1(Endothelin-1,ET-1)表达的影响。方法体外培养,利用高浓度葡萄糖诱导内皮细胞HUVEC-12细胞株功能损伤。实验分为正常对照组、低剂量高糖组、中剂量高糖组和高剂量高糖组,普通光镜和Hoechst 33258 荧光染色观察HUVEC-12细胞形态和核形态的变化;逆转录-聚合酶链式反应检测细胞内ET-1、vWF mRNA的表达;细胞免疫荧光检测细胞内ET-1、vWF蛋白水平的改变。结果与对照组细胞比较,不同浓度高糖处理内皮细胞72 h后,普通光学显微镜下观察到,细胞数目减少,圆形细胞明显增多,细胞间隙增宽,细胞边界不清;Hoechst33258染色结果表明有细胞核浓缩、出现高亮度蓝色荧光的凋亡细胞,并呈剂量依赖性损伤细胞;细胞ET-1、vWF mRNA及蛋白表达上调。结论高糖对人脐静脉内皮细胞有显著的损伤作用,可能与vWF、ET-1表达升高有关。 |

ObjectiveTo investigate the effects of human umbilical vein endothelial cells ET-1 and vWF expresstion induced by high glucose.MethodsHigh concentration of glucose induced endothelial cell dysfunction in HUVEC-12,and experiments were randomized into four groups:control group,low dose high glucose group,middle dose high glucose group and high dose glucose group.The change of cell shape and nuclear morphology were observed by light microscope and nuclear staining with Hoechst 33258.The mRNA expression of ET-1,vWF and GAPDH were assessed by reverse transcription PCR (RT-PCR).The protein content 1of ET-1,vWF were evaluated by cell immunofluorescence.ResultsCompared with the control group,the HUVECs incubated with different concentrations of high glucose (11.1 mmol/L,22.2 mmol/L,33.3 mmol/L),evidently showed damage.The results showed that after pretreatment with different concentrations of glucose for 72 h,the cells appeared typical characteristics of apoptosis including apoptotic nuclear condensation in the nuclear staining assay using Hoechst 33258,the expression of ET-1,vWF mRNA and protein were up-regulated.ConclusionsHigh concentration of glucose has a significant effect of HUVECs injury.The data suggest that ET-1,vWF may play an important role in the state of diabetic vascular complications. |
