| 目的探讨甲基化特异性聚合酶链法(MSP)和焦磷酸测序法(PS) 检测Runx 3基因甲基化的特异性。方法选取150例胃癌患者［依据美国癌症联合委员会(AJCC)分期标准，分肿瘤的早期，进展期两组］胃切除的胃癌标本及50例无癌变胃黏膜标本。分别以MSP法和PS法同时检测其Runx3启动子甲基化程度。结果胃癌组织与正常胃黏膜相比，其Runx3基因甲基化程度的差异均有显著性(MSP法：67.3%，40.0%，P=0.002；PS法：76.0%，30.0%，P<0.001)。MSP法显示：Runx3甲基化与肿瘤大小相关(P<0.05)；而PS法分析显示：进展期胃癌的Runx3基因甲基化率高于早期胃癌(P<0.05)；且其甲基化状态与肿瘤的大小(P=0.004)、Lauren分级(P=0.043)、浸润的程度(P<0.001)、淋巴结转移(P=0.021)及肿瘤TNM临床分期(P=0.045)等临床病理特征之间存在相关性。 |

| ObjectiveTo investigate the specificity of MSP and PS in detection of Runx3 gene methylation.MethodsWe selected 150 specimen from patients with gastric canner and 50 normal specimen in our experiment,and used MSP and ps tests to detect the methylation status of Runx3 gene promoter.ResultsCompared to normal tissue,there are dramatic difference in methylation status of Runx3 gene CpG island in human gastric cancer(MSP:67.3 vs 40.0%,P=0.002;ps:76.0 vs 30.0,P<0.001).MSP test demonstrated that Runx3 gene methylation was only related to tumor size(P<0.05).Ps test showed that the extent of Runx3 gene methylation was higher in advanced gastric cancer than in early gastric cancer,and the extent of methylation status of Runx3 gene was related to tumor size(P=0.04),Laurens classification(P=0.043),depth of invasion(P<0.001),lymph node metastasis(P=0.021),and TNM staging(P=0.045).ConclusionPS is more sensitive than MSP in detection of Runx3 gene methylation,and the methylation status of Runx3 gene detected by PS was closely related to clinicopathological characteristics of human gastric cancer. |
