目的比较人工培养的蛹虫草菌丝体与天然冬虫夏草抑制人胰腺癌PANC-1细胞株增殖的活性,并探讨其作用机制。方法MTT法分析蛹虫草菌丝体和天然冬虫夏草对PANC-1细胞的增殖抑制作用,Hoechst33258染色检测细胞凋亡,Western blot分析细胞凋亡和自噬相关蛋白变化。结果蛹虫草菌丝体和天然冬虫夏草都能通过诱导细胞自噬而非凋亡途径,呈浓度依赖地降低胰腺癌PANC-1细胞的存活率。癌基因STAT3的总蛋白和磷酸化水平均明显降低。蛹虫草菌丝体处理后PANC-1细胞促存活蛋白Mcl-1和Survivin的表达水平明显降低,而冬虫夏草并无此作用。结论人工培养的蛹虫草菌丝体与天然冬虫夏草相比,具有相似或更强的体外抗胰腺癌细胞增殖活性。 |

ObjectiveTo determine the anti-proliferative effects of cultured Cordyceps mycelium and natural Cordyceps sinensis on human pancreatic carcinoma PANC-1 cells and the possible mechanisms.MethodsThe reduction of cellular viability was determined by MTT assay.Hoechst 33258 staining was used to confirm the features of apoptosis.Western blot assays were performed to explore the apoptotic and autophagic pathway.ResultsBoth cultured Cordyceps mycelium and natural Cordyceps sinensis inhibited the proliferation of PANC-1 cells in dose-dependent manner via autophagic rather than apoptotic pathway.The expression and phosphorylation levels of STAT3 were deregulated remarkably.Treatment with cultured Cordyceps mycelium rather than natural Cordyceps sinensis obviously decreased the expression of Mcl-1 and Survivin in PANC-1 cells.ConclusionCultured Cordyceps mycelium has similar or stronger anti-proliferative activity than natural Cordyceps sinensis. |
