      目的探讨盐酸氮芥抑制人肝癌HepG2细胞生长的可能机制。方法以盐酸氮芥处理人肝癌HepG2细胞,运用DNA片断化分析、流式细胞术检测肝癌HepG2细胞的凋亡和细胞周期的改变。结果DNA片断化分析显示没有出现典型“梯形”条带,流式细胞术显示各浓度组细胞凋亡率之间差异无显著性(P>0.05),但有明显的细胞周期阻滞(S期和G2期)。结论盐酸氮芥对人肝癌HepG2细胞的生长抑制主要通过细胞周期的阻滞发挥作用。

      ObjectiveTo investigate the probable mechanism of nitrogen mustard to suppress human liver tumor HepG2 cells growth.MethodsThe apoptotic cells and the cell cycle were detected by DNA fragmentation analysis and flow cytometry.ResultsThe ladder-shaped strands werent observed from DNA fragmentation analysis;There was no significant difference in apoptotic rate among various concentration groups of nitrogen mustard by flow cytometry(P>0.05),but cell cycle arrest was distinctly observed at S phase and G2 phase.ConclusionsNitrogen mustard suppressed liver cancer HepG2 cells growth mostly by cell cycle arrest.
