目的研究三邻甲苯基磷酸酯(TOCP)对人成神经瘤细胞(SH-SY5Y)的氧化损伤作用。方法常规方法培养SH-SY5Y细胞,经TOCP染毒,分别检测不同浓度(0~1.0 mmol/L)TOCP作用48 h以及相同浓度TOCP(0.5 mmol/L)作用不同时间后(0、24、48、72 h)细胞内丙二醛(MDA)、过氧化氢酶(CAT)的含量变化,MDA含量用硫代巴比妥酸法(TBA)检测,CAT含量用可见光分光光度法检测。结果TOCP染毒SH-SY5Y细胞48 h的半数抑制浓度(IC50)为0.77 mmol/L。随着TOCP染毒浓度的增高,细胞内MDA的含量逐渐增高,CAT含量逐渐下降,这种变化趋势呈现明显的剂量—效应关系(P<0.05)。随着TOCP染毒时间的延长,细胞内MDA和CAT含量均呈明显的时间—效应关系。结论TOCP对SH-SY5Y细胞有明显的氧化损伤作用。 |

ObjectiveTo study the oxidative damage of tri-ortho-cresyl phosphate (TOCP) to human neuroblastoma SH-SY5Y cells.MethodThe cultured SH-SY5Y cells were treated with TOCP at different concentrations (0 ~1.0 mmol/L) or at the same concentration of TOCP (0.5 mmol/L) for different time (24,48,72 h),and then,intracellular malondialdehyde (MDA) and catalase (CAT) content were detected by thiobarbituric acid method (TBA) and visible light spectrophotometry respectively.ResultsThe half of inhibitory concentration (IC50) of TOCP treatment to SH-SY5Y cells for 48 h was 0.77 mmol/L.With the increased concentration of TOCP treatment,intracellular MDA content gradually increased,while CAT levels gradually decreased,which showed significant dose-response relationship (P<0.05).With the TOCP treatment time prolonged,intracellular MDA and CAT levels showed obvious time-effect relationship.ConclusionTOCP can cause oxidative damage on human neuroblastoma SH-SY5Y cells. |
