目的基于多次睡眠潜伏期测试方法,检测一种新颖的褪黑素受体激动剂Neu-P11对大鼠睡眠潜伏期和睡眠时间的影响。 方法大鼠多次睡眠潜伏期测试包括习惯化与检测两个阶段,两者间隔24 h。检测阶段包括总共3 h内的6个单个测试,单个测试时间是30 min;在每个单个测试中,先呈现5 min的干扰声刺激,之后25 min不呈现干扰声刺激。根据视频录像分析大鼠在25 min内入睡所需的时间(睡眠潜伏期)和睡眠时间。在检测之前6~8 min腹腔注射Neu-P11(5、10、20和50 mg/kg)或等体积溶媒。结果与溶媒组大鼠比较,Neu-P11(20和50 mg/kg)处理组大鼠其睡眠潜伏期显著性减少而睡眠时间显著性增加。结论在大鼠多次睡眠潜伏期测试中,Neu-P11可通过减少睡眠潜伏期增加睡眠时间,最终改善睡眠。 |

ObjectiveTo assess the effects of a novel melatonin agonist Neu-P11 on the sleep latency and sleep length in the rat multiple sleep latencies test(MSLT).MethodsThe MSLT included six separate sleep latency trails conducted within 3 h(30 min per trial).For each trail,the rats were initially kept awake for 5 min by means of gentle handling,and then were left undisturbed for 25 min.The time to fall asleep(sleep latency)and sleep length within the undisturbed 25 min period were recorded.The rMSLT was run 6~8 min after Neu-P11(5,10,20 and 50 mg/kg)was administrated.ResultsCompared with vehicle group,both the groups with Neu-P11 at 20 and 50 mg/kg showed a significant less level of sleep latency.At the same time,both the groups with Neu-P11 at 20 and 50 mg/kg,compared with vehicle group,showed a significant more level of sleep length.ConclusionNeu-P11 improves sleep by reducing sleep latency and increasing sleep length in the MSLT in rats. |
