      目的 探讨银杏叶提取物对大鼠脊髓全横断后神经营养因子表达的影响.方法 40只成年健康SD大鼠,随机均分为假手术对照组(SO组)、单纯脊髓横断组(tSCI组)、生理盐水治疗组(NS组)和银杏叶提取物治疗组(EGB组).每组分别于术后1天、7天各处死5只取材,检测神经元数量,并采用免疫组织化学ABC法,检测各组神经元细胞神经生长因子(NGF)和脑源性神经营养因子(BDNF)的表达.结果 SO组NGF和BDNF的表达较少,大鼠脊髓腹角神经元数量、NGF和BDNF的表达在24h和7天之间无变化;大鼠tSCI后,脊髓腹角神经元数量明显减少,NGF和BDNF的表达增加,损伤后时间越长,神经元数量减少越明显,NGF和BDNF阳性的细胞数增加越多;EGB治疗组脊髓腹角神经元数量较SO组减少(P<0.05),但较tSCI组、NS组增多(P<0.05),NGF和BDNF的表达较SO组、tSCI组和NS组增强(P<0.05).结论 EGB可使脊髓横断损伤后神经元数量增加,神经营养因子NGF、BDNF表达增加,EGB对SCI有保护和治疗作用可能与其对NGF、BDNF影响有关.

      Objective To explore effects of extract ginkgo bilobae leaves(EGB) on the expression of neurotrophins in transected spinal cord of rat.Methods 40 adult SD rats were randomly and averagely divided into 4 groups: SO group,tSCI group,NS group and EGB therapy group.Five rats in each point were sacrified at 24h or 7d after operation.The immunohistochemical ABC method was used to detect the expression changes of NGF and BDNF in each group and date of number of survival neurons.Results The number of neurons in the ventral horn of spinal cord in tSCI group had no apparent change compared with that of NS group at 24h and 7d(P>0.05),but that of EGB group increased more significantly than that of tSCI group and NS group.And the number of neurons in SO group was significantly higher than that of the above three groups at any time(P<0.05).The increasing expression of NGF and BDNF positive cells was observed 24h after operation in EGB therapy group,and reach peak value in 7d after operation,and that of EGB group were significantly higher than that of tSCI group and NS group(P<0.05).There was also difference between 24h and 7d in EGB group,tSCI group and NS group(P<0.05),but there was no difference between tSCI group and NS group at 24h and 7d(P>0.05).The expression of NGF and BDNF in SO group were significantly lower than that of the above three groups at any time(P<0.05),there was no change between at 24h and 7d in SO group.Conclusions EGB can significantly promote the expression of endogenous NGF and BDNF,and also maintain the survival of neurons in the ventral horn of spinal cord in following tSCI of rats.It provides a favorable condition for the self-plerosis or farther therapy of spinai cord following SCI.The mechanism of propection and therapy may be related to the increasing expression of endogenous NGF and BDNF.
