目的 观察罗哌卡因腹腔注射后对大鼠血液淋巴细胞DNA的影响,为不同浓度罗哌卡因的应用提供安全性依据.方法 Wistar大鼠72只,随机分为四组,对照组腹腔注射生理盐水,实验组(T1,T2,T3)按体重0.1 mL/100 g分别腹腔注射0.5%、0.75%、1.0%罗哌卡因.各组分别在腹腔注射后6、12和24小时3个时点各取6只动物,采用单细胞凝胶电泳检测血液淋巴细胞DNA损伤.结果 各实验组血淋巴细胞的彗星率和尾长均高于对照组,其中 T3组各时点与对照组相比差异有统计学意义(P<0.05);T2组与对照组相比,在24小时时差异有显著性(P<0.05).结论 低浓度(0.5%,0.75%)罗哌卡因腹腔注射24小时内对大鼠血液淋巴细胞无明显影响;高浓度罗哌卡因(1.0%)腹腔注射24小时内对大鼠血液淋巴细胞可能存在损伤作用. |

Objective To explore the lymphocyte DNA damages after celiac injection ropivacaine in wistar rats and provide the experimental data for the security assessment of ropivacaine application.Methods Seventy-two male wistar rats were randomly and averagely divided into 4 groups.The groups were randomized to receive normal saline,0.5%,0.75% or 1.0% respectively by the celiac injection 0.1 mL every 100 g body weight.Six rats in every group in 6 h,12 h and 24 h after the injection were killed,and every rat was phlebotomized blood approximately 0.5 mL blood(anticoagulant by heparitin).The alkaline single cell gel electrophoresis was used to determine DNA damages of blood lymphocyte.Results The comet tail ratio and tail length of lymphocyte in 1.0% concentration group at 6 h,12 h and 24 h were higher or longer than that of the control group at corresponding time point(P<0.05),while that of 0.75% ropivavaine group compared with the control was significantly different only in 24 h(P<0.05).The comet tail ratio and tail length of lymphocyte in 0.5% concentration group at 6 h,12 h and 24 h compared with the control had no significant difference(P>0.05).Conclusions 0.5% and 0.75% ropivavaine do not have obvious DNA damage effect on rats' lymphocyte.But 1% ropivavaine can induce DNA damages of rats lymphocyte,and it may have potential risk to human. |
