| 目的研究二烯丙基二硫诱导人髓性白血病HL-60细胞分化过程中蛋白质组的差异表达。方法形态学观察和硝基四氮唑蓝还原反应鉴定DADS诱导HL-60细胞分化；运用蛋白质组学方法（双向凝胶电泳、质谱分析、生物信息学方法）分析鉴定差异表达的蛋白质。结果DADS诱导HL-60细胞分化后，形态发生明显变化，NBT还原能力明显增强。通过质谱分析和数据库搜索初步鉴定了18个差异表达的蛋白质点，其中galectin-10在处理组中表达上调，钙网硬蛋白前体则表达下调。结论galeetin-10表达上调、钙网硬蛋白前体表达下调与DADS诱导HL-60细胞分化有关。 |

| Objective To study differential proteomic expression in the differentiation of HL-60 cells induced by diallyl disulfide(DADS).Methods Cell differentiation was validated by morphological method and nitroblue tetrazolium(NBT) reduction test.differential expression of cellular proteins were analyzed and identified via proteomic approach,two dimensional gel electrophoresis,mass spectroscopy and bioinformatics. Results The cytomorphosis of the DADS-treated cells was significantly different from controls.NBT reduction was increased in the DADS-treated cells(P<0.01).eighteen spots were identified by mass spectrometry which exhibited detectable quantitative changes between control and DADS-treated cells,unambiguously.Among of them,the expression of galectin-10 was increased in DADS-treated cells and the down-regulation was observed in the expression of calreticulin.Conclusion Up-regulated expression of galectin-10 and down-regulated expression of calreticulin were associated with HL-60 cells differentiation induced by DADS. |
