| 目的本研究应用蛋白质组学方法,分析胃癌(gastric carcinoma, GC)中表达缺失蛋白质分子.方法提取胃癌及配对正常胃黏膜组织总蛋白,采用双向电泳获得胃癌与配对正常胃黏膜组织蛋白质表达谱并进行分析,鉴定胃癌表达缺失的蛋白质分子.结果通过比较分析5对高分化胃腺癌及配对正常胃黏膜组织的蛋白表达谱,共发现差异表达蛋白点81个,其中7个蛋白点仅正常胃黏膜组织表达,而在胃癌组织中表达缺失,分别为:细胞周期素依赖性激酶抑制蛋白12(P16)、细胞周期素依赖激酶抑制蛋白1(P21)、抗氧化蛋白2、二硫化蛋白异构酶A2、C-1-四氢叶酸合成酶、Rho-GTPase-激活蛋白4、胰岛素样生长因子结合蛋白2.结论胃癌组织与正常胃黏膜组织之间存在蛋白质表达差异.寻找胃癌组织与正常胃黏膜组织之间表达差异蛋白质,为探讨胃癌发病分子机制提供有用依据. |

| Objective This study was designed to identify the differential proteins in gastric carcinoma (GC).Methods With two-dimensional electrophoresis (2-DE), protein profiles of normal and GC tissues were obtained. Proteins with altered expression were identified through matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS). Results Compared with the global protein patterns of normal gastric mucosa and GC, 81 differential protein spots were identified. Seven protein spots expressed in normal gastric mucosa, but disappeared in GC. They were cyclin-dependent kinase inhibitor 12(P16), cyclin-dependent kinase inhibitor 1(P21), antioxidant protein 2, protein disulfide isomerase A2, C-1tetrahydrofolate synthase, Rho-GTPase activating protein 4 and insulin-like growth factor binding protein 2. Conclusion There are differential expression proteins between normal gastric mucosa and GC. To search differential expression proteins between normal gastric mucosa and GC proves useful for further investigating the molecular mechanism of GC. |
