| 目的 探讨穿透支原体对宿主细胞的黏附机制。方法 用去污剂Triton X-100提取穿透支原体蛋白，用配体免疫印迹试验鉴定纤连蛋白结合蛋白(FnBP)；然后制备纤连蛋白Sepharose 4B亲和柱，运用亲和层析技术从制备的样品中分离纯化FnBP，在非还原状态下进行十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)。结果 配体免疫印迹试验鉴定出一65kDa的蛋白条带，可特异性地结合Fn；亲和层析技术得到相应的FnBP的纯化产物。结论 FnBP与穿透支原体对宿主细胞的黏附过程有关。 |

| Objective To study the adherence mechanism of Mycoplasma penetrans to host cells. Methods Mycoplasma penetrans proteins were extracted,and identified the FnBP by ligand immunoblot assay. Then the Fn coupled Sepharose was prepared, and the FnBP was purified by affinity chromatography technique. Results A protein of 65KDa which can bind Fn specifically was identified, and purified FnBP was obtained from Mpe proteins extract. Conclusion FnBP was corresponding to the adherence of Mycoplasma penetrans to host cells. |
