| 目的 探讨体外培养、扩增骨髓基质干细胞的方法，诱导骨髓基质干细胞向神经元样细胞定向分化。方法 采用全骨髓法培养成年SD大鼠股骨骨髓基质干细胞。传至第5代，用全反式维甲酸诱导分化，于诱导后第5小时行神经细胞特异性抗原MAP-2和胶质细胞特异性抗原GFAP鉴定。结果 成年鼠骨髓基质干细胞经全反式维甲酸诱导30min后可见神经元样细胞出现，经MAP-2免疫组化鉴定阳性。结论 (1)骨髓基质干细胞在体外扩增迅速，纯化需时短。(2)骨髓基质干细胞在全反式维甲酸诱导下向神经元样细胞分化。 |

| Objective To study the methods of culture and amplification of rat mesenchymal stem cells( MSCs) in vitro,and to induce MSC differentiating into the neuron-like cells. Methods The MSCs were isolated primarily from bone marrow of adult rat, and purified by passage culture. The 5th passage of MSCs were induced by all-trans-retinoic acid (RA) and immunocytochemical staining with anti-MAP-2, and GFAP were also performed on cells after five hours induction. Results Neuron-like cells appeared among MSCs after MSCs treated by RA. Neuron-like cells all were showed MAP-2 positive and GFAP negative. Conclusion MSC can be amplificated and purified in short time, be induced into neuronal-like cell by RA. |
