| 目的：研究胃癌的发生、发展及预后与p53基因突变的关系。方法：用多聚酶链反应和DNA双链直接测序方法进行p53基因外显子5、6、7、8序列分析，共检测33例胃癌、17例胃粘膜非典型增生、31例胃癌术后并转移性腹水标本。结果：发现9例胃癌和7例胃癌术后并发腹水标本存在p53基因碱基突变，突变率分别为27．27％和22．58％，且在7例腹水标本中发生p53基因突变的患者原病理切片组织中发现了相应的突变。17例胃粘膜非典型增生组织中亦发现1例p53基因突变。结论：胃癌与p53基因突变密切相关，p53基因突变是胃癌较早期的生物学行为。 |

| Objective: To study the action of p53 gene in the genesis, development and prognosis of gastric cancer. Methods: Polymerase chain reaction (PCR) and double-strand DNA sequencing were applied to analyse exons 5, 6, 7, 8 of p53 gene in 33 cases of gastric cancer,17 cases of gastric mucosal atypical hyperplasia and 31 cases of peritoneal effusion from patients with gastric cancer. Results:27.27%(9/13) cases of gastric cancer, 22.58%(7/31) cases of peritoneal effusion and 5.88%(1/17) case of atypical hyperplasia showed p53 gene mutations respectively.The mutations in peritoneal effusion were just the same as that in previous embeded tissues. Conclusion: There is a close relation between gastric cancer and p53 gene mutation which may be a early biological behaviour in gastric cancer genesis. |
