| 目的 建立对患者体内病毒水平进行客观定量检测的方法。方法 采用差示逆转录 -聚合酶链反应 ,共同扩增柯萨奇B组病毒 (CVB)目的基因和参照模板Mmyogene ,建立一种检测CVB复制水平的定量方法 ,并对苦瓜素体外抗CVB3感染疗效进行初步评价。结果 该方法成功检测到CVB3RNA水平 ,且苦瓜素处理组CVB3RNA水平明显低于对照组。结论 该方法具有简便、快速、可靠、重复性好等优点 ,同时检测多份样本 ,非常适合于大样本药物筛选 ,可为抗CVB病毒感染疗效的评价提供一个有效指标 |

| Objective:In order to evaluate the therapeutic effect,to screen effective drugs for CVB infective disease,and to observe the changes of patients, condition,it is necessary to develop an objective method to quantitate the levels of the virus. Method:Defferential RT PCR was used to co-amplify the target CVB RNA gene,to determine the level of CVB replication quantitatively and to evaluate the therapeutic effect of momordicin on CVB 3 infection in vitro. Result:The levels of CVB 3 RNA in myocardial cell treated with momordicin significantly lowered compared with those of the controls. Conclusion:This method is easy to control with the advantage of simplicity,reliability,quickness and reproducibility and provides an useful objective evidence for the evaluation of therapeutic effect on CVB infection. |
