      本文介绍一个不连续缓冲系的聚丙烯酰胺凝胶圆盘电泳和一个新的活性染色程序。用牛犊小肠和人胎盘硷性磷酸酶作内标和外标,分离鉴定人血清碱性磷酸酶同工酶,所得电泳图谱与萘基磷酸固兰 BB 盐染色改进法比较,清晰度、灵敏性、和重现性均较好,可以扫描定量。分析120例以上体检和肝病血清,各同工酶(肝、骨、肠和其它高分子量碱性磷酸酶四带)电泳迁移率与文献参考比值完全一致。

      This article aims to recommend a polyacryl amide gel disc electrophoresis with discontinuous buffer systems,and a newly defined Activity staining procedure,for separation and identifica- tion of alkaline phosphate isozymes in human serum,taking advantages of calf intestinal and human placental alkaline phosphatases either as internal or external standards.The isozymog- rams obtained by this method have good clarity,sensitivity,and reproduciability when compared with that of the currently available naphat- phosphate-fast blue BB salt staining,and Both can be quantitahyl tively scanned.Statistical treatment of the characteristic Rf values of each AP isoenzyme on the zymograms selected from more than 120 samples revealed very consistent results as those of the reference. Undoubtedly,this simple method will be benificial during ts application to clinical diagnosis.
