本文介绍一种制备抗 IgG 血清的方法,即利用含 A 蛋白的金黄色葡萄球菌(SPA 菌)吸附 IgG 的 FC 段后不改变 FC 段结构、仍保持其抗原特异性这一特点而制备抗血清.这样可省去提纯 IgG 的复杂步骤,同时 SPA 菌在免疫过程中又起到了良好的佐剂作用,增强了抗原的免疫原性。结果可获1:320的效价,免疫电泳鉴定纯度,仅在 IgG 图形部位出现一条粗沉淀线的抗 IgG 血清.此法简单,适用于教学实验. |

A simple easily adoptable method of preparing anti-IgG antibody is reported.The method is based on the principle that staphylococus producing protein A will absorb the FC fragment of IgG without alterring its structure and the antigenic specificity.Thus in preparing anti-IgG antibody the complicated procedures of purifying IgG can be omitted. Further more staphylococus producing protein A served as a good adjuvant enhancing the antigenicity of IgG in the process of immunization. The precipitating titer of the antibody reached 1:320 in electrophoresis with the antigen,a thick band of precipitation appeared in the region of IgG,confirming the purification of the antibody. |
