| 用硝酸银染色,并以100mmHg压力在体灌注固定液固定主动脉12小时,能较好地保持主动脉内皮完整。用该法制备的正常家兔主动脉扫描电镜样品,内膜表面光滑且平整,内皮细胞边界清晰可辨,细胞连接处的银线细小、均匀且连续。实验性动脉粥样硬化家兔主动脉内皮银染线明显增粗,形态僵硬,还可见双银染线及双核内皮细胞。静脉注射高脂血清兔主动脉内皮亦可见类似变化。 |

| After staining intercellular junction of endothelial cells with silver nitrate, the rabbit aorta was fixed by infusing fixative in situ at a pressure of 100 mm. Hg.for 12 hours. Using this method the morphological integrity of the endothelium was well maintained. Light microscopy of the silver stained aorta confirmed that silver salts were deposited at intercellular junctions. The intimal surface of the normal rabbit aorta examinedby SEM was even and smooth. The cell boundaries were clear. The cells were long-spindle or oval shaped and their long axis orientated in the direction of blood flow. The silver lines at intercellular junction were fine,uniform and continuous.The silver lines of the aortic endothelium of experimental atherosclerotic animal were shown to be increased in width obviousely and appeared to stiff. Double silver lines and binucleated cell were also seen. The similar changes were observed in the endothelium of the animal injected with hypercholesterolemie serum. In this article the technique was described in detail and its superiorities discussed. |
